Current Research on The Safety of Hormones & Psychedelics

When it comes to the complex interplay of hormones and tryptamine psychedelics, it’s important to first understand how psychedelics affect the brain, which we covered in a past article. 

Before we jump into the research, we’d like to point out that, similar to what was observed in cannabis, an “entourage effect” likely applies here as well, with psychedelics having a cumulative, synergistic effect on the human body.

In other words, depending on which psychedelic is being used, the unique combination of an individual’s hormonal makeup most likely affects how any given mind-altering substance manifests experientially. 

Currently, limited scientific research directly addresses the safety and potential risks of combining psychedelic therapy with HRT, so it is crucial to confer with a doctor or health professional before experimenting with potentially contraindicated substances. 

Firstly, serotonin, the well-known neurotransmitter related to the psychedelic experience, was even shown to act as a hormone in addition to its role as a neurotransmitter (Rybaczyk et al., 2005) in certain species.

Most importantly, it’s not fully understood just how important serotonin is in a wide variety of functions, but we know that it does play a role in cognition, mood, and behavior. 

Along with the effects of HRT on serotonin levels are estrogen, progesterone, and testosterone. 

According to a study from 1995, “estrogen could alter mood and mental state by increasing the density of 5-HT2A receptors in cerebral cortex and nucleus accumbens” (Sumner & Fink).

In another study from 2021 by Raval and colleagues, it was found that a single dose of psilocybin actually decreased 5HT2A receptor density, but increased synaptic density.

This strange effect on receptor density is likely related to the antidepressant abilities of entheogens like psilocybin mushrooms, however, it is not clear how an increase or decrease in receptor density would affect the safety of the experience. 

At the very least, it may have implications on the experienced potency of any given tryptamine psychedelic.  

In a 2000 study, the time it took for serotonin to bind to its receptors increased with the administration of estrogen and progesterone. This could suggest that HRT could lead to a delay in felt effects of serotonin agonists like psilocybin (Moses et al.).

The topic of hormones is truly complex and has greater implications on personality & spiritual transformation in relation to altering or treating personality and mood issues with hormones or psychedelics. 

While research on psychedelics is growing in numbers and all suggests efficacy for such issues, it has yet to extend to the topic of safety in those undergoing HRT.

Questions and considerations to be addressed in future research:

      • How does HRT affect the psychedelic experience directly? Do certain forms of HRT lead to greater sensitization to the effects of psychedelics
      • Are there contraindications between psychedelics like LSD, magic mushrooms (psilocybin), MDMA, DMT, etc. and hormonal treatments
      • Does increased synaptic density or decreased receptor density have any significant impacts on the efficacy of psychedelic therapy or the safety of it?
